Optimization of Growth Conditions to Enhance PHA Production by Cupriavidus necator

The accumulation of polyhydroxyalkanoates (PHAs) by microorganisms usually occurs in response to environmental stress conditions. Therefore, it is advantageous choose two-step cultivation. first phase aimed at maximizing biomass production, and only the second phase, after setting suitable conditions, PHA production starts. aim this work was optimize composition minimal propagation medium used for Cupriavidus necator DSM 545 using surface methodology (RSM). Based on results from search optimization limits, glucose concentration, ammonium sulfate concentration phosphate buffer molarity were chosen as independent variables. optimal values found follows: 10.8 g/L; 0.95 60.2 mmol/L. predicted 4.54 g/L, verified value 4.84 g/L. Although primarily focused determining medium, we also evaluated that 6.7 0.60 20 PHB yield 54.7% (w/w) dry biomass, 49.1%.
